PC gamers used to accessing the console for quick access to a number of nifty commands are going to be SOL when they realize they cannot do this in Duke Nukem Forever. Gearbox has removed the ability to access the console, and they explain why.
There has been a HUGE uproar over this on the Gearbox forums. Many had speculated this was to prevent changing of the FOV or modding the game in any other significant way for the single player experience.
Keep your fingers crossed that a patch is released soon that addresses this for PC gamers.

The console was disabled to prevent exploits and cheating in multiplayer. There were some very nasty things one could do in multiplayer with the console enabled and we couldn't in good conscience allow access to something that had the potential to spoil the multiplayer experience.
That said, we're examining ways to enable the console without impacting the multiplayer gameplay and I'll keep you posted.
