Then again, I don't speak for everyone. How about uploading 2-3 different versions? (Low-Quality, Medium and High.)
Let people know the file size for each version and then, depending on their download quotas, desire for image quality, etc. they can pursue the option that suits them best.
You could also consider using DivX or Xvid, you can keep it under 200mb for a 12min video and still have high quality. That's the encoding used in the Mine movies and a lot of popular Sir movies. I like using wmv since ppl don't have to download any codecs to watch the video, but it takes up a bigger filesize to get the optimal quality.
If you have 512kbps chances are it will take a long time to upload it and by the time it ends you won't want to consider re-encoding different versions. If I was in your position with a 300mb video I would encode with DivX at 1mbps - 2mbps bitrate at the highest quality encode, no matter the resolution, which will ensure less quality will be lost at the chosen bitrate (with the expense of encoding time.)
See if that helps reduce the size... remember, I have done a 32 minute video in 272mb and the quality was good enough.
Upload that 1 version to your non-streamable hosting site(s) Always upload to filefront no matter what people say. It's easier to have things stored and submitted from there then upload it directly and have it lost. (Remember, you can contact some hosts and have them grab the file using the fastpass account, so you won't have to upload again.) Upload the same version to google video too. The key being convenience, it will push people to download a better version if they really like it.
IMO, people won't care how big the video is if it's got the publicity. A highly anticipated and/or well marketed movies will always be downloaded. User reviews/feedback will also help post-release.
I would concentrate on screenshots and thread layout, during uploading time.
